Adsorption of norovirus and ostreid herpesvirus type 1 to polymer membranes for the development of passive samplers ArchiMer

Aims This study was performed to develop passive sampling methodology for the detection of two viruses in seawater in the area of shellfish production, The Norovirus (NoV), a human pathogen implicated in gastroenteritis outbreaks linked to oyster consumption and the ostreid herpesvirus type 1 (OsHV-1) a virus associated with mass mortalities of Pacific oysters. Methods and Results Commercially membranes were tested for their capacity to adsorb virus: Zetapor, gauze, nylon, low density polyethylene (LDPE) and Polyvinylidene difluoride (PVDF). The current study evaluates the effect of seawater physico-chemical characteristics on the relationship between the concentration of metals measured by Diffusive Gradients in Thin films (DGT) passive samplers (i.e., DGT-labile concentration) and the concentrations measured in discrete water samples. Accordingly, Inductively Coupled Plasma Mass Spectrometry (ICP-MS) was used to measure the total dissolved metal concentrations in the discrete water samples and the labile metal concentrations obtained by DGT samplers; additionally, lead and cadmium conditional labile fractions were determined by Anodic Stripping Voltammetry (ASV) and total dissolved nickel was measured by Cathodic Stripping Voltammetry (CSV). The Water European Framework Directive requires the implementation of programs to monitor the chemical quality of water bodies (freshwater, transition water and seawater), based on grab measurements of concentrations of various contaminants (metals, organic compounds). The results are limited by several analytical difficulties, and the poor representativity because of the temporal variability of contamination. The passive sampling techniques, which basically consist of concentrating substances on a submerged device for a given period, and analyzing accumulated substances, should improve the monitoring, by simplifying analytical issues (lower detection limits, analysis in a simpler matrix), and allowing time integration of the contamination. Use of low density polyethylene membranes for assessment of genotoxicity of PAHs in the Seine River ArchiMer

The genotoxicity of river water dissolved contaminants is usually estimated after grab sampling of river water. Water contamination can now be obtained with passive samplers that allow a time-integrated sampling of contaminants. Since it was verified that low density polyethylene membranes (LDPE) accumulate labile hydrophobic compounds, their use was proposed as a passive sampler. This study was designed to test the applicability of passive sampling for combined chemical and genotoxicity measurements. The LDPE extracts were tested with the umu test (TA1535/pSK1002 ± S9) and the Ames assay (TA98, TA100 and YG1041 ± S9).
